Manolis Kellis conducts research at the intersection of computational biology and genomics, with a primary focus on the systematic discovery of functional DNA elements. His highly cited work includes the development of ChromHMM for automating chromatin-state discovery, a method that has garnered over 2,000 citations for its role in characterizing genome-wide epigenetic patterns. Publicly indexed outputs suggest a long-standing interest in comparative genomics, evidenced by his early lead-author research into evolutionary analysis of ancient genome duplication in yeast and the identification of regulatory motifs through multi-mammal comparisons. His more recent senior-author contributions have translated these computational approaches into the biomedical sphere, notably through a landmark study on the circuitry of FTO obesity variants and adipocyte browning. Beyond his primary methodological developments, Kellis has contributed to large-scale international consortia aimed at mapping the human genome's functional landscape. He is a co-author on influential papers describing the GENCODE reference human genome annotation and the integrated encyclopedia of DNA elements. His collaborative work also extends to single-cell applications, including a highly cited transcriptomic analysis of Alzheimer’s disease that identifies cell-type-specific changes in the brain. These diverse outputs, ranging from the creation of the HaploReg tool to the study of immune bases in neurodegeneration, reflect a research arc dedicated to decoding the complex regulatory logic that links genetic variation to human disease.
