Research Statement

Daniel J. Drucker's indexed work centers on the biology and therapeutic translation of incretin hormones, particularly GLP-1 receptor signaling and its role in insulin secretion and glucose control. His early demonstration that glucagon-like peptide I stimulates insulin gene expression and raises cyclic AMP in a rat islet cell line helped establish the incretin field, while later reviews on the biology of incretin hormones and the mechanisms and therapeutic application of GLP-1 synthesized how these peptides act in type 2 diabetes. A senior-author study of glucose intolerance in mice lacking the GLP-1 receptor clarified the receptor's physiological necessity. Beyond these, his middle-author contributions include work on normalization of obesity-associated insulin resistance through immunotherapy and on FTO obesity variant circuitry and adipocyte browning in humans, reflecting broader metabolic and genetic interests. No software, datasets, patents, or grants are indexed beyond publications. Across these works, the throughline is a sustained effort to connect incretin hormone biology to clinical diabetes and metabolic disease, a line of inquiry shared with collaborators such as David A. D’Alessio.
