Barney S. Graham's most-cited work centers on structure-guided vaccine design for respiratory viruses. His senior-authored studies on stabilized prefusion RSV F glycoprotein trimers revealed how prefusion-specific antibodies drive RSV neutralization, while his later work on mRNA-1273 vaccine design enabled by prototype pathogen preparedness, with over 1,600 citations, linked coronavirus spike stabilization to rapid pandemic response. Related studies on heterosubtypic influenza protection via hemagglutinin-stem nanoparticles and DNA vaccine development for Zika virus extend this prefusion-antigen strategy across viral families, and his earlier investigations into RSV pathogenesis and T lymphocyte subsets established the immunological foundations for these approaches. Beyond these lead-author works, Graham contributed to foundational structural and clinical studies, including cryo-EM characterization of the SARS-CoV-2 spike in its prefusion conformation with Daniel Wrapp and Andrew B. Ward, and the phase 3 efficacy and safety evaluation of the mRNA-1273 vaccine. No software tools, datasets, patents, or funded grants are indexed beyond this publication record. Taken together, the named works trace a consistent arc: from defining RSV immunopathology and T-cell responses to engineering prefusion-stabilized antigens and shepherding them into clinical mRNA vaccine platforms.
