Ansuman T. Satpathy's research centers on the epigenomic and transcriptional programs that govern immune cell identity and dysfunction. His early work used Zbtb46 as a marker distinguishing classical dendritic cells from other lineages, and he later showed that Notch2-dependent classical dendritic cells orchestrate intestinal immunity to bacterial pathogens. A major thread concerns T cell exhaustion: he developed massively parallel single-cell chromatin profiling of immune development and intratumoral T cell exhaustion, and his laboratory has since combined genome-wide CRISPR screens identifying chromatin remodeling factors that limit T cell persistence with studies of divergent clonal differentiation trajectories underlying exhaustion. Beyond these, he contributed to an improved ATAC-seq protocol enabling interrogation of frozen tissues and to the chromatin accessibility landscape of primary human cancers, both widely used resources. His senior-author work includes single-cell analyses identifying brain mural cells expressing CD19 as potential off-tumor CAR-T targets and discovery of SARS-CoV-2 RNA-host protein interactions. Collaborations with Crystal Lee Mackall and Carl H. June connect this work to translational CAR-T efforts. Across these studies, his contributions trace a consistent arc: from defining immune lineages to mapping the epigenetic regulators that constrain T cell persistence and shape immunotherapy design.
